Microengineered heterogeneous substrates for cell culture by electro-microfluidics

摘要

Cultivating cells on a heterogeneous substrate prepared by assembly of hydrogels and arrangement of microparticles using electric manners on an electro-microfluidic (EMF) device are reported. We demonstrated a multifunctional integrated EMF device, combining (1) electrical manipulations of dielectrophoresis (DEP) and electrowetting-on-dielectric (EWOD) and (2) UV crosslinking technique, in order to manipulate, assemble, and fabricate complex microengineered hydrogels. With the ability to drive and assemble the hydrogel droplets and to arrange the microparticles within the hydrogel droplets, the highly programmable culture substrate provides an approach to tissue engineering, cell-cell interaction, stem cell differentiation studies. Poly(ethylene glycol) diacrylate (PEG-DA) and gelatin methacrylate (GelMA) were manipulated to form heterogeneous substrates for neonatal mouse cardiomyocytes culture.
